057 Targeting keratinocytes to potentiate skin immunization

Skin is a unique target for vaccination. Targeting keratinocytes to produce proinflammatory mediators improve immunity rational approach vaccine design. Keratinocytes genetically engineered overexpress the stress response related transcription factor x-box binding protein 1 (XBP1) increased production of and co-delivered antigen. Keratinocyte-specific overexpression XBP1 through cutaneous genetic immunization was transient induced pro-immunogenic skin microenvironment, including enhanced expression antigen, skin-infiltration stimulatory function dendritic cells. Overexpression in drove non-viral DNA vaccines enhance induction antigen-specific humoral cellular immune responses, durable skin-resident memory CD8 T cells effective protective mouse tumor models. These findings support strategy targeting immunization, identify as potential molecular ‘adjuvant’ contribute development efficacious skin-targeted against cancers infection diseases.
